General description
POLG is a mitochondrial protein that plays a major role in mitochondrial DNA replication and only for mitochondrial DNA. POLG is the catalytic subunit of a multiple protein complex that makes up the holo-enzyme that is the mitochondrial specific DNA polymerase gamma. POLG makes the alpha subunit of the complex. A number of diseases are associated with mutations that affect the function of POLG and the mitochondria including Leigh syndrome, various neurodegeneration diseases such as Parkinson's and Alzheimer′s, as well as various skeletal muscle and ophthalmoplegia disorders and even male infertility. The polymerase that POLG ultimately comprises is Polymerase gamma (Pol γ) and is the only DNA polymerase that is active in mitochondria and that can replicate mtDNA.
